The Physiological and Flavonoid Metabolism of the Tea Plant in Response to Nitrogen Nutrition is Regulated by the Ultraviolet-B Radiation Environment

摘要: The sensitivity of tea plants to environmental factors like UV-B is modulated by nutrients, subsequently leading to alterations in accumulation of quality components within tea plant. In order to elucidate the interactive impacts of UV-B radiation and nitrogen supply on the physiology and metabolism of tea plants, tea cuttings cultured via hydroponics were exposed to three distinct UV-B radiation durations and two nitrogen levels. The findings demonstrated that an extended UV-B radiation duration induced severe leaf structural damage, a build-up of reactive oxygen species, and photooxidative stress. Conversely, the activities of the enzymes associated with carbon assimilation and secondary metabolism declined, accompanied by a reduction in photosynthetic productivity and sugar content. Nevertheless, certain amino acids such as Ser, Glu, and Asp accumulated to relatively elevated levels under high UV-B (low nitrogen) conditions. Simultaneously, the expression of genes related to secondary metabolism, along with flavonoids, was upregulated. Deficient or excessive UV-B or high-nitrogen supplementation notably inhibited accumulation of most flavonol glycosides. Notably, a higher nitrogen level significantly promoted the accumulation of Quericetin-3-Glycoside and Myricetin-3-glucoside at a specific UV-B dosage, thereby maintaining a comparable total flavonol content in the high-nitrogen treatment to that in the low-nitrogen treatment. Our results suggested that diverse UV-B doses exert both physiological stress and metabolic regulation effects on tea plants, which might be correlated to antioxidation as a strategy for enhancing environmental adaptability of tea cultivar cuttings. These discoveries disclose the relationship between the tea plant's response to nutrient supply and light conditions, further providing a theoretical foundation for optimizing tea cultivate strategies.
