Antifungal activity and mechanism of volatile organic compounds produced by Bacillus velezensis strain P2-1 against Botryosphaeria dothidea-induced postharvest decay in apples

摘要: Apple ring rot caused by Botryosphaeria dothidea is among the most frequent postharvest diseases in apples during storage. This study aimed to investigate the biocontrol activity and potential control mechanism of volatile organic compounds (VOCs) produced by Bacillus velezensis strain P2–1 on B. dothidea-induced apple postharvest decay. The in vitro analyses indicated that VOCs produced by strain P2–1 markedly inhibited mycelial growth of B. dothidea strains and caused hyphal deformity and cell membrane destruction. Moreover, fumigation with the VOCs activated the expression of PR genes in apple fruit. In addition, VOCs treatment led to significantly changes in the richness, evenness, and diversity of bacterial communities and the richness of fungal communities on apple fruit surface. 13 specific VOCs in both B. velezensis strains P2–1 and G3–29 headspace mixtures were identified by headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) assay. The result of fumigation treatment showed that 6 of the 13 VOCs, including 2-octanol, acetic acid, 2-methylbutanoic acid, butanoic acid, isobutyric acid, and propanoic acid, exhibited strong antagonistic activity and caused hyphal degradation and cell membrane destruction in B. dothidea. Fumigation of apple fruit with each of the 6 VOCs strongly reduced the average diameter of B. dothidea-induced disease lesions, consistent with the result of treatment with strain P2–1 VOCs. Therefore, strain P2–1 VOCs and the 6 identified VOCs could serve as novel biocontrol agents against apple postharvest decay caused by B. dothidea.
