A haplotype-resolved genome assembly of tetraploid Medicago sativa ssp. falcata

摘要: High-yielding, high-quality and stress-tolerant forage crops are crucial for the sustainable development of grass-based livestock husbandry. Medicago species are some of the most important legume forage crops worldwide. Alfalfa (M. sativa subsp. sativa; M. sativa), a crop known as the “Queen of the Forages”, has high biomass productivity and nutritional value and good adaptability. M. sativa subsp. falcata (M. falcata) is closely related to M. sativa, and is widely distributed in cold regions of Russia, Mongolia, Scandinavia, and northern China, has outstanding tolerance to cold, drought, and soil infertility. M. falcata can be cross-pollinated with M. sativa to produce M. sativa subsp. varia (M. varia), which has better resistance and higher application value than M. sativa. Therefore M. falcata is an important genetic source for Medicago breeding. However, a reference genome of M. falcata has not been assembled until now. Here, using Illumina, PacBio HiFi, and Hi-C sequencing, we generated a high-quality, haplotype-resolved genome sequence assembly of an M. falcata accession originating from the former Soviet Union (Accession number: PI502453, PAVLOVSKAYA). This genome, which is highly continuous and complete, represents the first high-quality reference genome of M. falcata. This genome lays the foundation for a wide range of population genetics studies and experimental research in M. falcata. In addition, our study provides important germplasm resources and accelerates the genetic improvement of Medicago.
