Genome-wide identification and analysis of the abiotic stress responsiveness of the heat shock protein 90 gene family in Medicago sativa L

摘要: BackgroundHeat shock protein 90 (HSP90) is a highly conserved class of cellular chaperone proteins that are widely present in plants and are involved in the response to various biotic and abiotic stresses. The HSP90 gene has been extensively studied across numerous species, and several investigations on alfalfa (Medicago sativa L.) have demonstrated that HSP90 gene is implicated in plant-pathogen interactions. However, the HSP90 gene has not been fully identified in alfalfa.ResultsIn this study, the HSP90 genes in alfalfa were identified and characterized throughout the genome. A total of 29 MsHSP90s were identified from the alfalfa Xinjiangdaye genome and were categorized into three subgroups, I, II, and III, with 15, 9, and 5 members in each group, respectively. The MsHSP90 family members in each subgroup have similar intron and exon structures, and motif analysis revealed that the MsHSP90 gene family is highly conserved. Analysis of RNA-seq data of MsHSP90 genes in different tissues (roots, elongated stems, pre-elongated stems, leaves, flowers, and nodules) revealed that the expression of three MsHSP90 genes (MsHSP90-4, MsHSP90-14, MsHSP90-18) had tissue specificity and that 17 MsHSP90 genes were expressed in all six tissues, but their expression levels varied significantly. Analysis of MsHSP90 RNA-seq data under three stresses (salt, drought, cold) revealed that four MsHSP90 genes (MsHSP90-10, MsHSP90-11, MsHSP90-16, and MsHSP90-27) respond to all three stresses. In this study, the HSP90 gene family of alfalfa was comprehensively identified and analyzed, and the candidate genes related to adverse stress conditions were identified by analyzing RNA-seq data.ConclusionThe findings of this study provide critical information for further analysis of the HSP90 gene function in alfalfa, laying a foundation for enhancing its stress resistance.
