Genetic diversity, antibiotic resistance, and pathogenicity of Aeromonas veronii isolated from farmed largemouth bass (Micropterus salmoides) in the main aquaculture regions of China

摘要: Largemouth bass (Micropterus salmoides) is a vital freshwater aquaculture species in China but is susceptible to bacterial diseases which can result in huge economic losses. We performed an epidemiological survey of bacterial diseases in largemouth bass that covered Foshan, Wuhan, and Chizhou in China. A total of 39 strains were isolated from diseased fish; of these, 23 strains were highly homologous to Aeromonas veronii. These strains were classified into 13 sequence types (STs; 12 of which were novel STs) by multi-locus sequence typing (MLST); this depicted the significant inter-regional diversity of A. veronii. Drug resistance testing revealed that the isolated A. veronii strains exhibited multi-drug resistance with regional characteristics. All bacterial strains were resistant to vancomycin, penicillin, and clindamycin; and sensitive to cefuroxime and aztreonam. With regards to drugs approval for aquaculture, the isolated strains exhibited the highest resistance rate to enrofloxacin (87.0%) and the lower resistance rate (26.1%) to florfenicol. Zebrafish were infected with 13 representative A. veronii via immersion infection following dermal abrasion. This experiment led to identification of HZAU2283, a hyper-virulent strain. We sequenced the whole genome of the HZAU2283 strain and analyzed a range of virulence factors, including adherence factors, motility invasion adhesion factors, and secretory system factors such as type II secretion system and type III secretion system. Collectively, these data provide new insights into the regional prevalence, drug resistance, and virulence profiles of A. veronii and extend our understanding of the bacterial diseases that threaten the largemouth bass industry.
