Excessive humus fertilization accelerates antibiotic resistance genes dissemination in soil: Interspecies electron transfer-mediated ecological succession as a key mechanism

摘要: As a key mediator of interspecies electron transfer (IET), the role of humus in regulating the spread of antibiotic resistance genes (ARGs) remains unclear. Here, we investigated the fate of ARGs under different humus gradients and explored the underlying mechanisms of the IET regulation of ARGs dissemination. The results showed that IET increased the relative abundance of ARGs in soil by 20.95-23.20 %, with the most pronounced dissemination observed with high humus addition. In addition, IET reshaped the microbial community, significantly enhancing the ARG host microorganisms. Network analysis revealed that IET intensified interactions within the microbial community to promote horizontal gene transfer (HGT). Pseudomonas was recognized as the workhorse for the augmentation of IET-driven HGT. Moreover, niche calculations indicated that humus-mediated IET drove the succession of ARG host microorganisms towards specialist species with enhanced environmental fitness. This study enhances our understanding of how humus-driven microbial electrochemical processes influence ARG dissemination, with implications for soil amendment practices and biosafety in agriculture.
