Impact of common acaricide exposure of honey bee colonies for Varroa control on royal jelly metabolome and proteome

摘要: Royal jelly (RJ), a glandular secretion of nurse honey bees, serves as a human dietary supplement. While oral pesticide exposure has been found to compromise RJ quality, the impact of hive-applied acaricides for parasitic mite control remains unexplored. We employed integrated metabolomic and proteomic analyses to evaluate compositional changes in the RJ from honey bee colonies treated with four common acaricides (formic acid, oxalic acid, thymol, or amitraz) versus untreated controls. Principal component analysis of the time-course metabolome data showed distinct temporal segregation patterns rather than acaricide-specific clustering. The acaricide treatment induced minimal changes in metabolic composition, with only 5–14 out of 118 annotated metabolites demonstrating significant alterations in the oxalic acid-treated group. Notably, trans-10-hydroxy-2-decenoic acid (10-HDA), a critical indicator for RJ quality evaluation, remained unaffected by acaricide exposure. Proteomic analysis revealed no significant changes in the ten most abundant proteins (constituting 96.42 % of total protein abundance), except for decreased glucose oxidase levels in thymol-treated colonies. These findings indicate that routine acaricide application elicits minor alterations in the metabolomic and proteomic profiles of RJ, thereby preserving its premium-grade characteristics.
