Biomineralized Gd/Dy Composite Nanoparticles For Enhanced Tumor Photoablation With Precise T-1/T-2-Mr/Ct/Thermal Imaging Guidance

摘要: Nowadays, combined photothermal therapy (PTT) and imaging diagnosis based on nanotheranostic agents are attracting increasing interest in tumor therapy field. However, facile synthesis approach and excellent biocompatibility of the nanoagents are highly desired but still remain a challenge. It is also difficult to eliminate the phototoxicity of the agent to normal tissues resulting from the lack of tumor targeting by traditional approaches. Herein, a biosafe Gd-integrated Dy nanoagent (BSA:Gd,Dy@PDA-FA) was synthesized via a facile and environmental-friendly biomimetic strategy using BSA as template at physiological temperature, in which Gd and Dy elements were simultaneously doped for the first time into BSA system to realize multi-mode imaging. The as-prepared ultra-small NPs (13.9 nm) showed superior stability and excellent biocompability both in vitro and in vivo. Importantly, based on the doping of Gd/Dy and the modification with folic acid (FA), the NPs could effectively accumulate at tumor and achieve distinct contrast enhancement at tumor site for both T-1/T-2-MR/CT imaging in vivo. Because the existence of PDA, the NPs could completely ablate tumor, protect the tumor-bearing mice from tumor recurrence and inhibit lung metastasis under NIR laser. Our work highlight the feasibility of BSA-mediated biomineralization of a nanotheranostic agent, and suggest the attractive imaging guidance and effective tumor ablation property of the BSA:Gd,Dy@PDA-FA NPs.
