A multi-pronged approach to understanding the antifungal mechanism of chitooligosaccharide against Fusarium oxysporum, a major pathogen causing potato dry rot

摘要: Dry rot caused by the genus Fusarium is one of the main diseases in postharvest storage of potatoes, resulting in substantial economic losses. Environment-friendly biocontrol is emerging as a key research area for the prevention and control of potato dry rot. Chitooligosaccharide (COS) is a water-soluble amino-glucan with a low degree of polymerization and high inhibitory effect on some phytopathogens. However, the effect and mechanism of COS against Fusarium oxysporum have not yet been investigated. Thus, the objectives of the study were to investigate the effect and underlying mechanisms of antifungal property of COS against F. oxysporum using integrated physiological, cytological, and transcriptomic approaches. Our study showed that COS could inhibit mycelial growth, and spore germination, disrupt the chitin distribution in the cell wall and the integrity of the cell membrane, increase the sensitivity of F. oxysporum to the environment stressors such as high/low temperature and UV, and reduce the virulence of F. oxysporum. Transcriptome analysis of F. oxysporum with and without COS revealed a total of 3949 differentially expressed genes (DEGs), including 2293 were upregulated and 1656 were downregulated. The analysis of Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways and Gene Ontology (GO) terms showed that DEGs were mainly related to metabolism, transport, and localization. Overall, this study provides a novel strategy for developing COS as a potential biocontrol agent against potato dry rot.
