金钗石斛对MPTP诱导PD小鼠的神经保护作用及机制研究

摘要: 目的探究金钗石斛提取物(DendrobiumnobileLindl,DNL)在1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)诱导的小鼠帕金森病(Parkinsondisease,PD)模型中的神经保护作用。方法将小鼠随机分为对照组、模型组,美多芭组(阳性药组)、DNL(100、200、300mg/kg)组,通过腹腔注射(i.p)MPTP(30mg/(kg·d),7d)建立亚急性小鼠PD模型,进行步态分析实验、爬杆实验、悬挂实验等评估DNL对PD小鼠运动异常的影响;酶联免疫吸附测定法(ELISA)检测各组小鼠纹状体区和皮层区肿瘤坏死因子-α(tumornecrosisfactor-α,TNF-α),白细胞介素-6(interleukin-6,IL-6)、和白细胞介素-1β(interleukin-1β,IL-1β)表达的变化;蛋白质印迹法(Westernblot)检测各组小鼠纹状体caspase-3和caspase-9的表达情况。结果模型组与对照组相比,推进指数、支撑时相的步态指标有显著变化(P<0.05);与模型组相比,美多芭组、DNL(100、200、300mg/kg)组在左后、右前、右后推进指数,左后、右前、右后支撑时相均有明显改善(P<0.05);与对照组相比,模型组在爬杆上半段和总时间增加(P<0.01、P<0.001),调头时间和下半段时间无明显变化(P>0.05);与模型组相比,DNL(300mg/kg)组可以改善上半段时间(P<0.05),美多芭和DNL(100、200mg/kg)组无明显改善(P>0.05);与对照组相比,模型组的悬挂评分降低(P<0.05),与模型组相比,DNL(300mg/kg)可以增加悬挂评分(P<0.05),美多芭和DNL(100、200mg/kg)无明显改善(P>0.05);与对照组相比,模型组小鼠纹状体中TNF-α、IL-1β、IL-6含量增加(P<0.05),caspase-3和caspase-9蛋白表达水平显著增高(P<0.001);与模型组相比,美多芭、DNL(300mg/kg)组炎症因子指标均降低(P<0.05),皮层中各组含量无差异(P>0.05);caspase-3和caspase-9蛋白表达水平均降低(P<0.05)。结论DNL能够改善MPTP诱导的PD小鼠行为变化,减轻脑组织中的炎症反应,抑制神经细胞凋亡,在治疗PD药物及功能食品研发方面具有较好的潜力。
