Comprehensive screening of salt-tolerant rice germplasm using a fuzzy membership and PCA-based evaluation model

摘要: Soil salinization is intensifying globally, posing a serious threat to land resource utilization and agricultural productivity. Rice (Oryza sativa), a moderately salt-sensitive crop, is a preferred choice for reclaiming saline-alkali lands. Developing salt-tolerant rice varieties is therefore essential for enhancing soil productivity and ensuring global food security. Despite this urgency, comprehensive evaluation systems and reliable indicators for assessing rice salt tolerance remain limited. In this study, 631 rice germplasm resources were evaluated for salt tolerance and sensitivity at the seedling stage using a soil culture method. A robust evaluation model (D-value) was established by integrating correlation analysis, principal component analysis, and the fuzzy membership function method. Based on this model, the germplasms were classified into five categories: highly salt-tolerant (77), salt-tolerant (142), moderately salt-tolerant (144), salt-sensitive (158), and highly salt-sensitive (110). Key evaluation indicators, salt tolerance level, leaf mortality rate, and control shoot length were identified as effective predictors of salt tolerance. Interestingly, rapid seedling growth under control conditions showed a negative correlation with salt stress tolerance. These findings provide valuable theoretical insights and genetic resources for deciphering the molecular mechanisms of salt tolerance and for breeding salt-tolerant rice varieties, contributing to the sustainable use of saline-alkali farmlands and global food security.
